Research Interests
We are focused on the Fragment Based Lead Discovery (FBLD) targeting protein-protein interactions within the complex signal transduction network of tumor. We have accomplished the first industrial standard NMR Fragment Based Screening pipeline in China, covering from hit identification, structure-guided inhibitor evolution to cellular assays. The highly automated NMR fragment screening has been applied to a number of targets related to epigenetics and small GTPases. The high potent lead-like compounds will be employed as a small-molecule probe to protein-protein interaction network, which furthers our understanding of the molecular basis of tumor proliferation and metastasis.
We are also exploiting the application of NMR novel techniques, e.g., Residual Dipolar Couplings, Paramagnetic labelling and CEST, to large complex, e.g., nucleosome and its binding partner.
